What is PCR ?
Polymerase Chain Reaction (PCR) or the Nucleic Acid Amplification Test (NAT) in Canada is a molecular biological method for gene amplification in vitro. It makes possible to duplicate in large numbers (with a multiplication factor of the order of one billion) a known DNA or RNA sequence from a small quantity (of the order of a few picograms) of acid nucleic (specific DNA sequence (Amplicon)) and specific primers consisting of synthetic oligonucleotides of 20 to 25 nucleotides. It is thus possible, for example, to detect the presence of the HIV virus or to measure a viral load (concentration of the virus in the plasma), traces of GMOs (genetically modified organisms), or hepatitis B, C and D viruses. .
Increasingly used in forensic science, this technique is based on the combination of two factors:
- the enzymatic synthesis and specific double-stranded DNA specific initiation properties of thermostable DNA-dependent DNA polymerases;
- the hybridization and dehybridization properties of complementary strands of DNA as a function of temperature.
These elements make it possible to control the enzymatic activity by means of temperature transitions (performed by a thermocycler) repeated cyclically.
